Derivation This line was produced by fusing Y3/Ag1.2.3 (ATCC CRL-1631) cells with spleen cells from an AO strain rat. Comments Tested and found negative for ectromelia virus (mousepox). The cells do not secrete either heavy or light immunoglobulin chains. Complete Growth Medium The base medium for this cell line is ATCC-formulated Dulbecco's Modified Eagle's Medium, Catalog No. 30-2002. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%. Subculturing Cultures are maintained by addition or replacement of fresh medium. Start new cultures at 1 X 105 viable cells/mL and maintain at or below 1 X 106 cells/mL. Medium Renewal: Every 2 to 3 days Cryopreservation Complete growth medium 95%; DMSO 5% (v/v). Cell culture tested DMSO is available as ATCC® Catalog No. 4-X.
