Applications
The line was produced by fusing a G-418 resistant derivative of the U-266 human myeloma cell line with P3X63Ag8.653 mouse myeloma cells.
The cells are useful as fusion partners for human B cells in the generation of human hybridomas.

G-418 resistance was introduced to the human myeloma by transfection with pSV-2 neo r.
The cells are useful as fusion partners for human B cells in the generation of human hybridomas.
Complete Growth Medium The base medium for this cell line is ATCC-formulated Iscove's Modified Dulbecco's Medium, Catalog No. 30-2005. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 20%.
Subculturing
Protocol: Cultures can be maintained by addition of fresh medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 2 x10(5) viable cells/ml. Maintain cultures at a cell concentration between 1 x 10(5) and 1 x 10(6) cells/ml. Scrape attached cells if necessary.
Medium Renewal: Every 2 to 3 days

Freeze medium: 75% CompleteGrowth Medium + 20% FBS + 5% DMSO
