ATCC determines the biosafety level of a material based on our risk assessment as guided by the current edition of Biosafety in Microbiological and Biomedical Laboratories (BMBL), U.S. Department of Health and Human Services. It is your responsibility to understand the hazards associated with the material per your organization’s policies and procedures as well as any other applicable regulations as enforced by your local or national agencies.

ATCC highly recommends that appropriate personal protective equipment is always used when handling vials. For cultures that require storage in liquid nitrogen, it is important to note that some vials may leak when submersed in liquid nitrogen and will slowly fill with liquid nitrogen. Upon thawing, the conversion of the liquid nitrogen back to its gas phase may result in the vial exploding or blowing off its cap with dangerous force creating flying debris. Unless necessary, ATCC recommends that these cultures be stored in the vapor phase of liquid nitrogen rather than submersed in liquid nitrogen.

Handling procedure
1. Open vial according to the enclosed instructions.

2. Using a glass vessel containing 50 ml of the recommended broth, withdraw 0.5 to 1.0 ml and rehydrate the pellet. 

3. Incubate the tube at 30°C, and incubate the glass vessel on a reciprocal shaker at the same 
temperature.

4. After 24 hours, transfer 1 mL of the culture into a fresh tube of broth.
This tube will be held in the refrigerator to be used as a host stock, if needed. 
It will also contain the Bdellovibrio, but often a more concentrated suspension of 
the host is needed for infection.

A pure culture of the host strain, ATCC 15144 Escherichia coli, is available and may 
also be used instead of trying to propagate the host as a mixed culture.

5. After several days, examine a wet mount preparation of the glass vessel culture to check for free swimming Bdellovibrio cells. They will be observed as very tiny, actively motile, curved rods.

Handling notes
A glass vessel must be used for propagation, not a plastic vessel.
Observe the flask culture microscopically daily until the Bdellovibrio are observed.  A decrease in turbidity should also be noted indicating lysis of the host cells. This may take 3-4 weeks for the host to be fully lysed.
To further propagate the strain, use the tube of the host set aside for this purpose.  It should be transferred and used as a 24 hour culture.
