ATCC highly recommends that appropriate personal protective equipment is always used when handling vials. For cultures that require storage in liquid nitrogen, it is important to note that some vials may leak when submersed in liquid nitrogen and will slowly fill with liquid nitrogen. Upon thawing, the conversion of the liquid nitrogen back to its gas phase may result in the vial exploding or blowing off its cap with dangerous force creating flying debris. Unless necessary, ATCC recommends that these cultures be stored in the vapor phase of liquid nitrogen rather than submersed in liquid nitrogen.

1. Pre-chill media by placing at 4°C for 30 minutes.
2. Open vial according to enclosed instructions.
3. Using a single tube of #2 broth (5 to 6 ml), withdraw approximately 0.5 to 1.0 ml with a Pasteur or 1.0 ml pipette. Rehydrate the entire pellet.
4. Aseptically transfer this aliquot back into the broth tube. Mix well.
5. Incubate the broth tube for 48 to 72 hours at 18°C.
6. Transfers can now be made to other tubes of broth, slants, and/or plates. This strain needs to establish good growth in the broth tube before additional passages are made. Additional incubation may be required for growth on solid medium.

To check for luminescence, inoculate a slant leaving cap loose. Incubate at 18oC until growth is established. Then, hold in the dark for 10 minutes before checking. If no luminescence is detected, reincubate and check regularly until luminescence appears. When maximum luminescence is obtained, stopper tightly.
Colonies on #2 agar are irregular, flat, translucent, circular, smooth, and yellowish.
